Title Transcriptome profile of drug resistant metaplastic breast cancer cell lines

Overall design Cells were treated with increasing concentrations of drug until they were able to survive and proliferate after a pulse of cytotoxic drug. HS-TX: HS578T cells, 100 nM paclitaxel, 5 days pulse; HS-DOX: HS-578T cells, 100 nM doxorubicin, 3 days pulse; BAS-TX: BAS cells, 20 nM paclitaxe, 3 days pulse; BAS-DOX, BAS cells, 160 nM doxorubicin, 3 days pulse. For the array experiment, RNA isolated from three (drug resistant cells) or four (parental naive cells) independent cultures were used for each condition.
Summary Two metaplastic breast cancer cell lines were used: a widely available, HS578T, and a novel line isolated from a metaplastic breast cancer tumor, BAS. Doxorubicin and paclitaxel resistant derivatives of these lines were generated and transcriptome profiling performed.
